Anti?Inflammatory Effects of Quercetin on High?Glucose and Pro?Inflammatory Cytokine Challenged Vascular Endothelial Cell Metabolism

Scope Pro-inflammatory stimuli such as hyperglycemia and cytokines have been shown to negatively affect endothelial cell functions. The aim of this study is assess the potential quercetin its human metabolites overcome deleterious effects hyperglycemic or inflammatory conditions on vascular endothelium by modulating metabolism. Methods results A metabolomics approach enabled identification quantification 27 umbilical vein (HUVEC) metabolites. Treatment HUVECs with high-glucose concentrations causes significant increases in lactate glutamate concentrations. Quercetin inhibits glucose-induced adenosine 5?-triphosphate (ATP) also increased inosine Tumor necrosis factor ?-treatment (TNF?) asparagine decreases aspartate Co-treatment reduces pyruvate compared TNF?-only treated controls. Subsequently, it was that HUVEC phase-2 conjugates inhibit deaminase, xanthine oxidase 5?nucleotidase (CD73) but not ectonucleoside triphosphate diphosphohydrolase-1 (CD39) purine nucleoside phosphorylase activities. Conclusion alter balance towards a less inflamed phenotype, both alone presence pro-inflammatory stimuli. These changes are consistent inhibition particular enzymes involved metabolism
